Design: The a1 and a2 haploid cells were isolated from single tetrads using micromanipulation. DNA was extracted using a Carver hydraulic press and the Qiagen Anion-exchange columns. Haploid genomes were sequenced using the P6/C4 Pacific Biosciences SMRT technology
